The Proof-of-Concept project aims to develop a vaccine against severe gastric disease and cancer.
Our recent results suggest that duodenal ulcer disease and gastric cancer can be caused by insufficient immune responses of blocking antibodies against the adherent gastric onco-pathogen Helicobacter pylori with consequential increased and high mucosal inflammatory pressure. We can show immune response with such blocking antibodies by vaccination in primates and mice.
And we can show full protection by vaccination with H. pylori attachment proteins against gastric cancer development in our mouse model. In the Proof-of-Concept project we will develop an mRNA Vaccine.
We will develop an mRNA Vaccine that will generate the necessary H. pylori attachment protein antigens for immune response with blocking antibodies.
The read-out will be protection against gastric cancer in our unique Leb-mouse gastric cancer model, with a humanized ABO antigen glycosylation pattern in the stomach mucosa. We coordinate the biological-functional investigation with analyses by radio immuno assays, Biacore and calorimetry. We have full access to state-of-the-art microscopy at the BICU/NMI.
Our new results open-up for novel use and faster development time-line for therapeutic vaccination against gastric disease. An mRNA Vaccine could reduce the risk of fatal bleeding ulcers and the risk of gastric cancer.
